The Histone Methyltransferase SETDB2 Regulates Abdominal Aortic Aneurysm Formation

چکیده

The purpose of this project was to establish whether the histone methyltransferase, SETDB2, regulates abdominal aortic aneurysm (AAA) development. We hypothesized that during AAA development, SETDB2 is upregulated, resulting in “turning off” genes are normally protective against pathologic vascular remodeling via methylation H3K9 at gene promoters. Specifically, normal wall existence, a balance exists between matrix metalloproteinases (MMPs), which function break down wall, and tissue inhibitors MMPs (TIMPs), prevent actions on wall. During an imbalance occurs TIMPs, favor MMP activity. Thereby, we Results from Setdb2 TIMP expression, causing proceed unregulated lead destruction C57BL/6 Setdb2f/fLyzCre+ mice, have macrophages deficient underwent angiotensin-II infusion murine model were compared littermate controls with intact expression (C57BL/6 wild type). Aneurysm size measured ultrasound after 28 days infusion. In vivo then isolated analyzed for activity promoter sites TIMPs 1, 2, 3, 4 using chromatin immunoprecipitation. mice had significant reduction formation their wild-type littermates (Fig 1). Furthermore, repression promoters significantly reduced 2). Taken together, these suggest epigenetic macrophages, leading wall.Fig 2H3K9me3 (TIMPs) littermates. collected weeks (n = 6 mice/group).View Large Image Figure ViewerDownload Hi-res image Download (PPT)

Oncogenic roles of the SETDB2 histone methyltransferase in gastric cancer

SETDB2 is a histone H3 lysine 9 (H3K9) tri-methyltransferase that is involved in transcriptional gene silencing. Since it is still unknown whether SETDB2 is linked to carcinogenesis, we studied alterations and functions of SETDB2 in human gastric cancers (GCs). SETDB2 protein was highly expressed in 30 of 72 (41.7%) primary GC tissues compared with their normal counterparts by immunohistochemis...

Drug Therapy for Small Abdominal Aortic Aneurysm

Dear Editor,Abdominal aortic aneurysm is often asymptomatic, less recognized, and causes considerable mortality and morbidity, if missed. The incidence varies from country to country and the occurrence is influenced by modifiable (smoking, coronary heart disease, hypertension, dyslipidemia, and prolonged steroid therapy) and non-modifiable risk factors (increasing age, male gender, and positive...

KLF4 regulates abdominal aortic aneurysm morphology and deletion attenuates aneurysm formation.

BACKGROUND KLF4 mediates inflammatory responses after vascular injury/disease; however, the role of KLF4 in abdominal aortic aneurysms (AAAs) remains unknown. The goals of the present study were to (1) determine the role of KLF4 in experimental AAA; and (2) determine the effect of KLF4 on smooth muscle (SM) cells in AAAs. METHODS AND RESULTS KLF4 expression progressively increased at days 3, ...

The Paraoxonase Gene Cluster Protects Against Abdominal Aortic Aneurysm Formation.

OBJECTIVE Abdominal aortic aneurysm (AAA) is a life-threatening vascular pathology, the pathogenesis of which is closely related to oxidative stress. However, an effective pharmaceutical treatment is lacking because the exact cause of AAA remains unknown. Here, we aimed at delineating the role of the paraoxonases (PONs) gene cluster (PC), which prevents atherosclerosis through the detoxificatio...

Membrane-Bound Thrombomodulin Regulates Macrophage Inflammation in Abdominal Aortic Aneurysm.

OBJECTIVE Thrombomodulin (TM), a glycoprotein constitutively expressed in the endothelium, is well known for its anticoagulant and anti-inflammatory properties. Paradoxically, we recently found that monocytic membrane-bound TM (ie, endogenous TM expression in monocytes) triggers lipopolysaccharide- and gram-negative bacteria-induced inflammatory responses. However, the significance of membrane-...
